Results: We tested the efficacy of P元7 at two different phases of this preclinical model. Methods: This study used von Frey filaments to measure periorbital mechanical thresholds, and grimace scores were evaluated by observing mouse facial expressions. We have shown previously that repetitive restraint stress (two hours per day, three consecutive days) induces periorbital hypersensitivity and facial grimace responses as well as priming to the nitric oxide donor sodium nitroprusside (SNP 0.1 mg/kg). To evaluate whether the endogenous opioid system could potentially be utilized as therapeutic approach for migraine, we tested the efficacy of the Dual ENKephalinase Inhibitor (DENKI) (P元7) in a preclinical model of migraine. One sentence summary: Inhibiting enkephalinase in the periphery effectively treats migraine-like behavior mainly via activation of delta-opioid receptors.īackground: Opioids are commonly used as second-line migraine treatment in the emergency department, but their use can lead to medication overuse headache or transformation from episodic to chronic migraine. 5ġ University of Texas at Dallas, Richardson, TX, USA 2 Pharmaleads, USA 3 Pharmaleads, Paris, FRA 4 University of Texas at Dallas, Richardson, TX, USA 5 University of Texas at Dallas, Richardson, TX, USA CATEGORY: BASIC SCIENCE OR-10 Efficacy of a dual ENKephalinase inhibitor in a preclinical stress-induced migraine model
